How does Criptine 2.5mg Tablet work?

Criptine works by activating dopamine receptors and reducing prolactin secretion from the anterior pituitary gland, which can help with conditions like galactorrhoea, infertility, and acromegaly by lowering growth hormone levels.

Side Effects of Criptine 2.5mg Tablet

• Common: Nausea, headache, dizziness, fatigue, lightheadedness, vomiting, abdominal cramps, nasal congestion, constipation, diarrhea, drowsiness.
• Hypotensive Effect: Possible slight hypotensive effect.
• Laboratory Abnormalities: Transient increases in blood urea nitrogen, SGOT, SGPT, GGPT, CPK, alkaline phosphatase, and uric acid.

Quick Suggestions:

• Use Criptine for conditions caused by excess prolactin, such as infertility or abnormal breast milk production.
• Take it with or after food to avoid side effects like nausea.
• Rise slowly from sitting or lying down to prevent dizziness or fainting.
• Avoid driving if you feel dizzy or sleepy after taking Criptine.
• Do not use Criptine if breastfeeding.
• Regular blood pressure checks may be necessary during treatment.

Indication of Criptine 2.5mg Tablet

1. Hyperprolactinemia-Associated Dysfunctions: Used for conditions associated with hyperprolactinemia, including amenorrhea with or without galactorrhea, infertility, or hypogonadism.
2. Prolactin Secreting Adenomas: Pre-surgery treatment to reduce tumor mass.
3. Acromegaly: Used to manage elevated growth hormone levels.
4. Parkinson's Disease: For idiopathic or postencephalitic Parkinson's disease, as an adjunct to levodopa (with or without a peripheral decarboxylase inhibitor).

Pharmacology

Bromocriptine mesylate is a dopamine receptor agonist that primarily inhibits the secretion of prolactin by activating post-synaptic dopamine receptors. In acromegaly, it lowers elevated blood levels of growth hormone. It significantly reduces plasma levels of prolactin in patients with both physiologically elevated prolactin and hyperprolactinemia.

Dosage & Administration of Criptine 2.5mg Tablet

• General: This should be taken with food. Evaluate frequently during dose escalation to determine the lowest effective dose.
• Hyperprolactinemic Indications: Initial dose: 0.5 to 2.5 mg daily, with gradual increments of 2.5 mg every 2-7 days until optimal response. In children aged 11-15: initial dose 0.5 to 2.5 mg daily, increasing as tolerated to a therapeutic range of 2.5-10 mg daily.
• Acromegaly: Start with 0.5 to 2.5 mg at bedtime with food for 3 days, increasing every 3-7 days until optimal benefit is achieved. Max dose: 100 mg/day.
• Parkinson's Disease: Start with 0.5 of a 2.5 mg tablet twice daily with meals, increasing every 14-28 days by 2.5 mg/day as needed. Max dose: 100 mg/day.

Interaction

• Decreased Efficacy: Dopamine antagonists, butyrophenones, phenothiazines, haloperidol, metoclopramide, pimozide.
• Not Recommended: Concurrent use with other ergot alkaloids.

Contraindications

• Uncontrolled hypertension.
• Sensitivity to ergot alkaloids.
• Discontinue use for hyperprolactinemia when pregnancy is diagnosed.
• Postpartum period in women with a history of coronary artery disease and other severe cardiovascular conditions.

Pregnancy & Lactation

• Pregnancy Category B: Use cautiously during pregnancy.
• Lactation: Not recommended for postpartum women during lactation.

Precautions & Warnings

• Renal or Hepatic Disease: Safety and efficacy not established; use with caution.
• Blood Pressure: Monitor if using other medications that lower blood pressure.
• History of Psychosis or Cardiovascular Disease: Use with caution.
